If I were to go to calculated shipping – domestic only – and a buyer were to purchase 2 or more items, will the weight total up and then calculate shipping from that new weight?

That is what will happen. If you have entered the weight of the individual items into the listing, then the calculator should total the items and get a total weight for the buyer’s order. To allow for the packaging materials, you may need to add the weight of the materials (padded envelopes, boxes, etc.) to the weight of the item so that the weight in the listing reflects the packaging materials.
I wouldn’t worry about being too exact, since anything over 13 ounces is usually going to be shipped via Priority Mail, UPS, or FedEX and they all round up to the next pound anyway.
